I don't recall the plane, and haven't seen the movie, but judging from the era, and Hughes' history, I would imagine it being a Grumman Goose. It mostly depends on how accurate the film-makers wanted to be on a historical basis, but considering the numbers of amphib planes out there these days, I would go with the Goose.

If you ever get the opportunity to go to Oregon, make sure to swing by McMinnville and the Evergreen Museum. The impressiveness of the Spruce Goose makes the trip worthwhile. The films of it flying don't do it the justice it deserves. It absolutely dwarfs a B-17 parked under its tail.
