I am curious if it would be possible to run a 4-ohm sub (single voice coil) from both sets of positive and negative on my amp. Is this possible and would I get more from my amp (mono class D) if I did this?

Just use one set of terminals. You will not gain anymore power by using both setts. As Sparky said, those terminals are just an extra set for adding more speakers. Convenience is the only reason for the other set.

Your amp will be seeing a 4 ohm load, and thus producing the wattage specified @ 4 ohms. Add another 4 ohm sub to the other set of terminals, and you will then have a 2 ohm load. Then your amp will theoretically be producing double the power, but usually only 85-90% more power with a mid-level amp. If you want it louder, add another 4 ohm sub and you will probably gain another 6 dB.
there are several ways to hook up the speakers to a amp. there is mono , stereo , and bridged modes. all car audio sub are gonna be 4 ohms.if you want power from both sides of the amp then i suggest you put into bridged mode.

To tell you the truth, I'd never done that. I suggest maybe getting a 2 ch bridgeable amp for the power. note: Make sure the RMS matches from the amp and sub.Actually you can't bridge a mono amp.

I'm not so sure that Lucky has installed many amps as the "all car audio sub are gonna be 4 ohms." is false. Subs can come in a variety of impedances.
