Need some clarification on what exactly you are looking for. Where exactly are looking for a voltage drop at?The coil is a step up transformer, so its output is going to be many times greater than the voltage input. Resistance the primary measurements taken on coils to see if they are operating within spec.
As others have said you'd normally test the resistance of both windings.

The only voltage drop I can think of in the circuit on some vehicles is the ballast resistor. Is this possibly what you want to test ?
You would again normally test it's resistance but you may want to test the voltage supplied to the coil via it.
During normal running the coil should be connected via the ballast, generally at around 9v depending on the car. When cranking it's bypassed so the coil gets the full available battery voltage.
