Have 2000 ram 1500 p/u w/ code P0601 and It's not shifting?

Question:
HAS USED TRANNY, OLD ONE BURNT OUT
ALSO CHANGED THE OUTPUT SPEED SENSOR
AND TRIED CHANGING THE PCM

That's a pcm problem, and you've changed out the pcm already, there may be an electrical spike going to the pcm, so check all wires and connections going to the brain to make sure there's no shorts. Also, with a used tranny, if you have a lift kit on the truck ( I have an 01 1500 with a 4" lift and 315's), make sure you turn the overdrive off when going under speeds of 55. It will chew up the tranny by constantly changing gears in overdrive mode. Even if the tranny was new, make sure to do that. I went through two already because of it. It may be your used tranny to begin with, the relay in the a/t is probably not functioning properly, and it may be caused by the electrical spike going to the pcm. Don't bother going to the dealer or shops to clear the code, it won't clear and they'll just tell you to change the pcm.

Automatic trannys bolted up to gasoline engines, need vacume to shift. look and be sure all hoses that carry vacume, are in their correct places. some ware on that tranny is a vacume nipple, intended for a vacume hose to be connected to it.

Is it bad to by a car that has had a new transmission?

if it is bolted to a diesel engine,there is no available vacume for it, and has a sensor system to make it shift

This is not for the amateur to diagnose take it to a professional you are way in over your head. If you listen to people that tell you this transmission is vacuum controlled you are in trouble. That guy has no clue what he is talking about!
